Formulation and Evaluation of Gastroretentive Floating Beads of Valsartan

Background: This study focuses on the formulation and optimization of floating calcium pectinate beads encapsulating valsartan, a BCS Class II drug with poor aqueous solubility and low oral bioavailability. Method: Floating drug delivery systems were developed using the inotropic gelation technique, incorporating low methoxy pectin (primary polymer), gellan gum (release modifier),
sodium bicarbonate (gas-generating agent), and calcium chloride (cross-linking agent). A 2³ factorial design was employed to optimize three independent variables: the ratio of pectin to gellan gum, sodium bicarbonate concentration, and calcium chloride concentration, with the goal of achieving prolonged gastric retention, high drug entrapment efficiency, and controlled drug release.
